چکیده

the present work with consideratlon to the autoradiographic pictures, suggests that cholinergic receptors are located at the gate of a channel originating from synaptic cleft coming to lie within the muscle fibre. ache molecules stand at the gate of this channel, controlling the entrance of different cholinergic agents. it was report- ••• ed previously that dtc molecules s t.abd ;:.2e the ache rnolecules and will obstruct the gate. this blocks the acess of ionic flux within the channel thus producing a non-depolarizing neuromuscular paralysis.the presented experiments imply that depolarizing agent will bring a considerable change in conformation of ache mole cule and this causes the opening of the gate allowing ioni flux and depolarization .in case of ach this process is repeated in a fraction of milli second, due to rapid regeneration of ache while in case of suxamethonium and neostigmine(given in high dose), the regeneration of ache takes much longer time thus will produce a depolarizing blockade. in this hypothepis the main responsa~ility of ache is confined to identification of cholinergic agents and cooperation in their function so,it can be accepted as cholinergic receptor. in regard to clinic, this work suggests that only the use of minimum effective dose of neostigmine is advisable, in reversing curarisation. in contrast to general belief , the dose of neostigmine should be s elec t ed in relation to r eceptor dtc occupation and not depending on pati ent 's weight . as it was demonstrated , the early use of high dose o f neostigmine may a lso potent i a te curar i s a tion

The Ubiquity Nature of Acetylcholine

Acetylcholine (ACh) was viewed as the “vagusstoff” after Loewi`s experiment with the frog heart [1]. In 1936 Otto Loewi, and Henry Dale were recipients of the Nobel Prize in Physiology and Medicine, by their contribution for the acetylcholine discover as a neurotransmitter [1]. So, ACh was regarded as a neurotransmitter during several decades [2]. After that it was evidenced a widespread expres...

Demonstration of the transmembrane nature of the acetylcholine receptor by labeling with anti-receptor antibodies

Antibodies raised in rabbits to Triton-solubilized, purified acetylcholine receptor from Torpedo californica were used to immunospecifically label intact T. californica electroplaque membrane vesicles attached to cover slips and oriented with the extracellular face of the synaptic membrane facing outward. Hemocyanin conjugated to Protein A was then used as a marker, making the antibody binding ...

Conformation of acetylcholine bound to the nicotinic acetylcholine receptor.

We report here the biologically active conformation of acetylcholine when bound to the high-affinity state of the receptor from Torpedo californica. The acetylcholine conformation was determined in the free and bound states by proton NMR two-dimensional nuclear Overhauser effects. In agreement with x-ray crystallographic data, acetylcholine in solution has an extended conformation with an avera...

Exploring the nature of molecular recognition in nicotinic acetylcholine receptors.

Nicotinic acetylcholine receptors (nAChRs) are the subject of ever increasing interest because of their presumed involvement in the etiology of numerous clinical disorders. Unfortunately, the absence of atomic-level structural data, as well as the pharmacological complexity of these receptors leaves many fundamental questions unanswered. An understanding of how ligands interact with the recepto...

The molecular neurobiology of the acetylcholine receptor.

Introduction The acetylcholine receptor (AChR) is the most thoroughly characterized component of the neuromuscular transduction process. Earlier reviews that summarize the structural and biochemical features of the AChR include Popot & Changeux (1984), Stroud (1983), Conti-Tronconi & Raftery (1982), and Karlin (1980). This receptor translates the binding of the neurotransmitter, acetylcholine (...
